Noncovalent complexes of the enzyme leucine aminopeptidase (LAP) with a variety of flavonoids have been characterized in solution by spectroscopic methods. Flavonoids are known to interact with biological targets by hydrophobic forces, but their mode of binding to the enzyme is not yet completely understood. We have used fluorescence titration to characterize the binding affinity of the flavonoids epigallocatechin gallate (EGCg), epigallocatechin (EGC) and epicatechin gallate (ECg) to LAP. The flavonoids were found to bind to LAP in the micromolar range. From the analysis of the stability constants of the complexes, it is evident that the complexes formed with EGCg and ECg are more stable than the complex with EGC. Isobestic points of the titration curves in both cases were observed, suggesting the formation of a 1:1 complex. The formation constants obtained were used to evaluate the contribution of the nonpolar interactions in the binding of flavonoids to LAP. The role of the hydrophobic interactions in the complex stabilization is discussed. a dark blue and a dark red shade.
